Boxes and cellophane are vapor-permeable. That means air - and water vapor - can and do pass through them. Both ways.

That said, Spanish cedar plywood (what most cigar boxes are made out of) and cellophane will slow down the exchange of water between the cigars and the ambient atmosphere. Additionally, since wood is hygroscopic, it can form something of a buffer against rapid RH fluctuations. Cellophane will protect the cigar's wrapper from the kind of damage you might get from handling the cigar as you go rooting around in the humidor trying to find that particular smoke you're looking for.

Bottom line: I never remove cellophane from a cigar until I plan to smoke it, and I age my cigars in their boxes.
